Santiago werd kort na Baracoa gesticht en werd al snel de belangrijkste stad van Cuba, vóór de oprichting van Havana. Van aanzienlijk historisch belang, Santiago was het toneel van een van de belangrijkste veldslagen tijdens de Spaans-Amerikaanse oorlog in 1898. Deze strijd markeerde het begin van het Cubaanse onafhankelijkheidsproces. Later, in 1953, markeerde de aanval op de Moncada-kazerne het begin van de Cubaanse revolutie.
